| I. Background | |
| CD16A（FcγRIIIA）是低親和力 IgG Fc 受體，屬免疫球蛋白超家族成員，主要表達于自然殺傷（NK）細胞、單核細胞及巨噬細胞表面。其核心功能為介導抗體依賴性細胞介導的細胞毒性（ADCC），通過識別抗體（如 IgG1/IgG3）的 Fc 段觸發(fā) NK 細胞殺傷腫瘤細胞，并參與免疫復合物清除、炎癥介質(zhì)釋放及細胞因子調(diào)控。 基因存在關(guān)鍵多態(tài)性位點，如第 158 位氨基酸的 V/F 變異：V 型（纈氨酸）與 Fc 段親和力強，F(xiàn) 型（苯丙氨酸）親和力弱，直接影響抗體治療療效（如曲妥珠單抗在乳腺癌中的反應）。此外，CD16A 的 N-糖基化修飾及金屬蛋白酶 ADAM17 介導的脫落（近膜區(qū)剪切）調(diào)控其功能，脫落的 sCD16 競爭性抑制 ADCC，成為腫瘤免疫逃逸機制。 |
